This thesis describes the design of a convenient format of solid phase microextraction (SPME) for bioanalysis in pharmaceutical industry and the validation of the approach to the application. An automated in-tip SPME technique coupled with liquid chromatography (LC) and tandem mass spectrometry (MS/MS) for high throughout drug analysis has been developed and applied to the quantitative determination of various drug compounds in different biological fluids from drug discovery to clinical development. The initial research in this thesis focused on a proof-of-concept study using manual multi-fiber approach to determine a drug compound in human plasma from a clinical trial. The proof-of-concept was achieved based on the validation data and a head-to-head comparison with conventional liquid-liquid extraction (LLE) method. An in-tip SPME technique was then proposed to explore the feasibility of SPME automation and two approaches of preparing in-tip SPME fibers were developed including fiber-packed and sorbent-packed fiber preparation. A simple and high throughput in-tip SPME fiber fabricating procedure based on polymer monoliths using photo-polymerization was introduced to prepare 96 fibers simultaneously. The biggest advantage of the in-tip SPME technique is that it is simple and easy to use for automation without introducing any additional devices and in the meantime, the simplicity of SPME is maintained. Automated in-tip SPME was applied to routine drug analysis in drug discovery and development environment. One case study involved the determination of vitamin D3 in human serum with derivatization and the in-tip SPME approach was compared with traditional LLE method using either tubes or 96-well plate extraction. Another study was to use hydrophilic interaction chromatography (HILIC) -MS/MS to determine three polar compounds, imipenem (IMP), cliastatin (CIL) and [beta]-lactamase inhibitor (BLI) simultaneously in different biological fluids including rat plasma and mouse blood. The results from both studies clearly demonstrated that in-tip SPME could be used as an alternative sample preparation method in bioanalytical analysis. Matrix effects in bioanalysis using automated in-tip SPME and LC-MS/MS were then thoroughly evaluated for the first time. Our study indicated that the assumption that SPME should provide sample clean up as effective as or better than solid phase extraction (SPE) with no or minimal matrix effects might not be always true, and matrix effects should be investigated in any SPME assays in bioanalysis.

Bioanalysis has gained importance in the scientific at an exponential rate. The focus of these works attempt to seek new approaches towards faster and more efficient bioanalysis that require little to no sample preparation. One approach is the use of new restricted access media trap technologies to facilitate an online sample preparation platform. A demonstration of the advantages of RAM involves the trace level detection of bisphenol A, an environmental contaminant. A bulk derivatization strategy used in combination with RAM trap technologies is adopted to boost sensitivity without the use of traditional sample preparation methods. Another strategy is the use of ambient ionization techniques. Novel affinity mesh screen materials are used to facilitate rapid drug discovery. The combination of these novel affinity materials with TM-DESI allows for the eventual creation of a truly high throughput screening process for new antibiotic drug compounds. A new ambient ionization technique, continuous flow - extractive desorption electrospray ionization is introduced in the presented work. Charge state manipulation of protein charge states are performed using CF-EDESI. The CF-EDESI technique is also coupled to high pressure liquid chromatography for a powerful analytical method with great versatility.

The area of biosample analysis encompasses a very broad range of assays which support the clinical and nonclinical studies. Biosample analysis is used to provide a quantitative or qualitative measure of the active drug and/or its metabolite(s) in the biological matrix for the purpose of pharmacokinetics, toxicokinetics, bioequivalence, and exposure-response (pharmacokinetics /pharmacodynamics) studies. Due to the significance of pharmacological analysis, sensitive, reproducible and robust analytical methods are critically needed for pharmacological studies of the biosamples. A bioanalytical method mainly contains two components I) Sample preparation II) detection of the compound. Therefore, the main aims of this thesis are the development of quantitative and qualitative analytical methods for the target compounds using LC-MS(/MS) and development of accelerated sample preparation for high throughput sample analysis for DNA and proteins.In this dissertation, a brief review on the method rationale, workflow of the method development, sample preparation methods, instrumentations and analytical method validation, are discussed in Chapter 1. Also, research projects were discussed and the techniques used in the experiments for this thesis were reviewed. As so, chapter II and III were mainly focused on the accelerated sample preparation methods for the high throughput sample analysis of DNA and proteins respectively, where the sample preparation time was significantly reduced from hours to minutes, which are suitable for qualitative and quantitative analysis of DNA and proteins. In Chapter IV, a systematic study on the structural characterization of the model glycoprotein Human IgG was described. In chapter V successful development of LC-MS method was developed for the determination of Oxygen -18 isotope enrichment in the phosphate samples in the positional isotope exchange reactions to study the reversibility of certain enzymatic reactions was described. Successful development and validation of a new and sensitive analytical LC-MS/MS method for the determination and quantitation of incorporation rates of decitabine, an anti-cancer drug which can be applied to determine the sensitivity and responsiveness in patients treated with decitabine was described in Chapter VI.
