The third wave of PFAL business started in around 2010 in Japan and Taiwan, and in USA and Europe it began in about 2013 after the rapid advances in LED technology. Plant gene transfer achieved in the early ‘80s paved the way for the exploitation of the potential of gene engineering to add novel agronomic traits and/or to design plants as factories for high added value molecules. For this latter area of research, the term "Molecular Farming" was coined in reference to agricultural applications in that major crops like maize and tobacco were originally used basically for pharma applications. The concept of the “green biofactory” implies different advantages over the typical cell factories based on animal cell or microbial cultures already when considering the investment and managing costs of fermenters. Although yield, stability, and quality of the molecules may vary among different heterologous systems and plants are competitive on a case-to-case basis, still the “plant factory” attracts scientists and technologists for the challenging features of low production cost, product safety and easy scale up. Once engineered, a plant is among the cheapest and easiest eukaryotic system to be bred with simple know-how, using nutrients, water and light. Molecules that are currently being produced in plants vary from industrial and pharmaceutical proteins, including medical diagnostics proteins and vaccine antigens, to nutritional supplements such as vitamins, carbohydrates and biopolymers. Convergence among disciplines as distant as plant physiology and pharmacology and, more recently, as omic sciences, bioinformatics and nanotechnology, increases the options of research on the plant cell factory. “Farming for Pharming” biologics and small-molecule medicines is a challenging area of plant biotechnology that may break the limits of current standard production technologies. The recent success on Ebola fighting with plant-made antibodies put a spotlight on the enormous potential of next generation herbal medicines made especially in the name of the guiding principle of reduction of costs, hence reduction of disparities of health rights and as a tool to guarantee adequate health protection in developing countries.

We are facing global issues concerning environmental pollution and shortages of food, feed, phytomass (plant biomass) and natural resources, which will become more serious in the forthcoming decades. To solve these issues, immeasurable numbers of various plants and huge amounts of phytomass are required every year for food, feed and for the improvement of amenities, the environment and our quality of life. Increased phytomass is also required as alternative raw material for producing bio-energy, biodegradable plastics and many other plant-originated industrial products. Only by using phytomass as a reproducible energy source and raw material, instead of fossil fuels and atomic power, we can save natural resources and minimize environmental pollution. To increase phytomass globally, we need billions of quality transplants (small plants) to be grown yearly, in the field or in the greenhouse, under various environmental conditions. However, these high quality transplants can be produced only under carefully controlled, rather than variable environment al conditions. Recent research has shown that the closed transplant production system requires considerably small amounts of electricity, water, fertilizer, CO) and pesticide to produce value-added transplants as scheduled with minimum release of environmental pollutants and minimum loss of transplants. The closed or closed-type transplant production system is defined as a transplant production system covered with opaque walls with minimized or controlled ventilation rates, using artificial lighting. With this system, photoperiod, light intensity and quality, air temperature, humidity, CO) concentration and air current speed can be controlled as desired.

This book provides two basic concepts on plant propagation and value-added transplant production in a closed structure with artificial lighting: 1) photoautotrophic (sugar-free medium, photosynthetic or inorganic nutrition) micropropagation systems, and 2) closed transplant production systems with minimum resource consumption and environmental pollution. This book also describes the methodology, technology and practical techniques employed in both systems, which have been commercialized recently in some Asian countries such as China and Japan. We often use a closed structure such as a tissue culture vessel, a culture room, a growth chamber, a plant factory with lamps, and a greenhouse to propagate plants and produce transplants. Main reasons why we use such a closed structure is: 1) higher controllability of the environment for desired plant growth, 2) easier protection of plants from damage by harsh physical environment, pathogens, insects, animals, etc, 3) easier reduction in resource consumption for environmental control and protection, and 4) higher quality and productivity of plants at a lower cost, compared with the plant propagation and transplant production under rain, wind and sunlight shelters and in the open fields. Thus, there should be some knowledge, discipline, methodology, technology and problems to be solved on plant propagation and transplant production common to those closed structures, regardless of the types and sizes of the closed structure.
